Elecoglipron is an orally administered, small-molecule agonist of the glucagon-like peptide-1 (GLP-1) receptor currently under investigation for the management of type 2 diabetes. The SOLSTICE phase 2b trial evaluated the efficacy, safety, and tolerability of once-daily elecoglipron compared with placebo. This randomised, double-blind, placebo-controlled study was conducted across medical research centres and hospitals in nine countries. Eligible participants were adults aged 18 years or older with a body-mass index of at least 23 kg m-2 and type 2 diabetes (glycated haemoglobin [HbA1c] 7.0-10.5%) managed with diet and exercise alone or monotherapy with metformin or a sodium-glucose cotransporter-2 inhibitor. At week 26, elecoglipron reduced HbA1c in a dose-dependent manner relative to placebo, with the highest-dose group achieving a placebo-adjusted reduction of approximately 1.5 percentage points. Weight loss was observed across all elecoglipron dose groups. The safety profile was consistent with the established GLP-1 receptor agonist class, with gastrointestinal adverse events as the predominant tolerability signal. These results support the progression of elecoglipron into phase 3 evaluation as a convenient, restriction-free oral option for glycaemic control and weight management in type 2 diabetes.
