Type 2 diabetes mellitus affects over 500 million adults worldwide (ref). Injectable GLP-1 receptor agonists such as semaglutide have demonstrated robust glycaemic control and weight loss, but the requirement for subcutaneous administration represents a barrier to treatment adherence (ref). Elecoglipron is an orally administered, small-molecule GLP-1 receptor agonist designed for once-daily dosing without food or fluid restrictions. Here we report the results of SOLSTICE, a phase 2b, randomised, double-blind, placebo-controlled trial evaluating the efficacy, safety, and tolerability of elecoglipron across a range of doses in adults with type 2 diabetes. This trial provides dose-ranging evidence to inform phase 3 development of the first orally administered, restriction-free GLP-1 receptor agonist.
